Unbiased analysis of antibody responses to all human viruses using Infinity Bio’s VirSIGHT library

Methodology and validation for pan-viral antibody reactome profiling at 100% sensitivity versus orthogonal reference assays.

VirSIGHT is Infinity Bio’s pan-viral antibody reactome assay, designed to detect antibody binding against every known human virus from a single serum or plasma sample. Built on Infinity Bio’s in-solution MIPSA (Molecular Indexing of Proteins by Self-Assembly) chemistry, the library encodes 285,669 unique peptides derived from over 2 million viral protein sequences spanning 500+ viral species and 94,000+ taxonomic IDs. This white paper documents analytical performance against three benchmark cohorts (SARS-CoV-2, HIV-1, and HSV1) characterized by orthogonal LFIA, NAT, and CLIA reference assays, and benchmarks VirSIGHT head-to-head against PhIP-Seq on identical peptide sequences. For groups designing longitudinal cohort studies, cross-sectional disease-association studies, vaccine programs, or surveillance and pandemic-preparedness work, the paper makes the case that comprehensive unbiased pan-viral profiling reveals associations single-pathogen serology cannot.

  • Library design: MSA-tiled ~60 amino-acid peptides with 20 aa overlap compress 2M+ UniProt viral sequences into 285,669 informative DNA-barcoded peptides covering 500+ species and 94,000+ taxonomic IDs.
  • Analytical validation: 100% sensitivity against SARS-CoV-2, HIV-1, and HSV1 reference cohorts, with no false-positive VARscores in seronegative samples and ROC-derived cutoffs of 0.71, 0.17, and 3.6.
  • Reproducibility: mean total-hits CV 7.08% and mean Pearson R 0.989 across plates and operators in 6× replicate; top-50 peptide FOB CV 20.4% across three independent MIPSA library lots.
  • VirSIGHT versus PhIP-Seq head-to-head: on identical peptide sequences and informatics, MIPSA produced 10–100× fewer false positives for off-cohort viruses such as Ebola and Rabies.
