What the World Put In Me (EnviroSIGHT)

A Southern soul ballad about the environmental record a body has been keeping since before it had a name, and what it takes to decipher it.

The exposures that shape an immune response arrive without announcing themselves. Pollen, mold, the microbial cargo of one particular stretch of road: each of them met an antibody response, and that response is still in circulation years after the encounter. EnviroSIGHT™ is Infinity Bio’s environmental exposure antibody reactome profiler, the world’s largest panel of allergens, non-viral and microbial antigens. One serum sample, screened against all of them in a single experiment.

Genomics is the sequence a person is born with. EnviroSIGHT is the record of everything after. Antibodies raised against an exposure keep circulating long after the encounter, so what accumulates is a historical archive, shaped by the order in which a lifetime of exposures actually happened. Where other omic layers describe the body’s intrinsic state, the antibody reactome describes what it responded to. Antibody Reactomics is the field built on deciphering that layer.

One serum sample, one experiment, and the libraries can be combined.

MIPSA is the technology that deciphers it, opening non-viral trigger discovery at a scale not previously possible. Antigens are expressed cell-free and coupled to DNA barcodes, and after immunoprecipitation with the sample’s antibodies those barcodes are counted by sequencing. Each catalog library is built on a non-overlapping barcode set, so more than one can be combined in a single immunoprecipitation reaction to profile antibodies of the same isotype. A study that wants the environmental record alongside the viral or the human one gets both. One sample, one reaction.

What EnviroSIGHT covers:

  • 31,133 unique protein antigens and allergens, tiled into 242,884 peptides of roughly 90 amino acids.
  • 1,207 genera and 3,437 unique taxonomic identifiers.
  • One microliter-scale serum sample, screened against all of them in a single experiment.
  • Non-overlapping barcode sets, so EnviroSIGHT can be combined with the other catalog libraries in one reaction, profiling antibodies of the same isotype.

The exposure does not have to be named in advance to be found.

A panel this size is what makes hypothesis-free discovery possible. Reactivity profiles across the library can be compared between groups and associated with the outcome under study, which is how an environmental trigger becomes a candidate instead of a hypothesis someone already held. Infinity Bio runs EnviroSIGHT as a research service, and the questions put to it are questions of etiology and biomarker discovery: which exposures associate with which phenotypes.
